Rampant Hearts powered eight ahead of Celtic at the summit of the Scottish top flight to strengthen their early title credentials and worsen the struggles around the Celtic team.
A superb goal from Kyziridis and a spot-kick from Shankland gave the Hearts team a 3-1 lead that the reigning champions could not recover from.
Dane Murray - among three youth players in the Celtic starting lineup - slashed into his own net in the early stages to put Hearts ahead at a energetic Tynecastle, and it was the youngster's infringement that led to the penalty.
Callum McGregor's classy first goal of the season had quickly cancelled out the first goal, and Benjamin Nygren almost put Celtic in front, choosing placement over power and allowing Alexander Schwolow to make a crucial stop.
But the unstoppable Hearts side regained dominance to hand Celtic to back-to-back losses for the initial occasion since last year.
It also gives the Hearts a commanding advantage at the summit.
The previous occasion they were this far ahead was in 2005-06 under George Burley, when they ultimately finished second - one of just four occasions that a team has failed to win the title after building such an advantage in the past 27 years.
The Hearts coach, whose Aberdeen side were second to Celtic on multiple times, has been working hard to play down title talk, as the club seek their maiden league title since 1960.
The Celtic boss has experienced this previously at the club - both flawless and stuttering starts to a season. He's yet to finish second best.
But on Sunday's evidence, this Hearts team showed they deserve serious consideration.
Analysis: The hosts Take Advantage of Depleted The Visitors
The Edinburgh side started with the confidence of a side undefeated in the league this season - fans' favourite Kyziridis poked a cross in for Shankland to fire off target.
The Glasgow team didn't help themselves with some unusual sloppiness in possession and they buckled under their initial test.
A headed effort from Braga was tipped onto the woodwork by Kasper Schmeichel and Shankland's return ball was turned into the net by the unfortunate Murray, in for the absent center-back.
The away side grew into the contest and started building moves. From one such move, the midfielder released the full-back to set up McGregor, who slotted home.
Apart from the Nygren chance, Celtic struggled to create much for striker Kenny or his substitute Yamada amid the unavailability of stricken attackers Kelechi Iheanacho and Daizen Maeda.
The home team rediscovered their energy and adaptability in creating chances in the opposition half. The Greek forward, a standout performer of the club's scouting network, fired a low shot past the goalkeeper from the edge of the box.
Immediately after, Murray fouled the midfielder in the right side of the box and Shankland converted. And it could've been an even more handsome win, had replacement Ageu not been denied by Schmeichel.
The visitors' performance after the break was partially encapsulated in two incidents involving record signing Arne Engles.
The set-piece from Engles delivery was in stark contrast to his midweek exploits in the Europa League win over their Austrian opponents and the national team player was then booked for a cynical foul.

The Hearts manager: "You see how hard the players work, the team spirit. That's not always enough to win games consistently.
"Our strength lies in, is skillful players. You're seeing that from multiple individuals who have been here before. Some of the new signings are keen to demonstrate their quality.
"We're in a positive moment at the minute, but it's only three points. It's crucial for us to maintain this form. We've got much work ahead, plenty to improve. Following the initial fixtures is normally an indication of where you are."
Celtic manager Brendan Rodgers: "It's a really poor goal from our perspective. We didn't move when the play was recycled as a line so when they pass the ball, they should have players offside but didn't. We have got to do better with the attempt.
"There's still so many points to play for. It's only the beginning. It's just nine matches. Clearly, being eight adrift is not ideal. The positive is there's so many games to play.
"Our attention must turn on the present and look to achieve greater stability and more quality in our play."

The Glasgow club are back at home on midweek against promoted side Falkirk (19:45 GMT). At the same time in the Scottish Premiership, the Edinburgh outfit are at their opponents' ground.
